چکیده

The goal of this review is to present a comprehensive survey of the many intriguing facets of creatine (Cr) and creatinine metabolism, encompassing the pathways and regulation of Cr biosynthesis and degradation, species and tissue distribution of the enzymes and metabolites involved, and of the inherent implications for physiology and human pathology. Very recently, a series of new discoveries have been made that are bound to have distinguished implications for bioenergetics, physiology, human pathology, and clinical diagnosis and that suggest that deregulation of the creatine kinase (CK) system is associated with a variety of diseases. Disturbances of the CK system have been observed in muscle, brain, cardiac, and renal diseases as well as in cancer. On the other hand, Cr and Cr analogs such as cyclocreatine were found to have antitumor, antiviral, and antidiabetic effects and to protect tissues from hypoxic, ischemic, neurodegenerative, or muscle damage. Oral Cr ingestion is used in sports as an ergogenic aid, and some data suggest that Cr and creatinine may be precursors of food mutagens and uremic toxins. These findings are discussed in depth, the interrelationships are outlined, and all is put into a broader context to provide a more detailed understanding of the biological functions of Cr and of the CK system.

Cli. the Metabolism of Creatine Ii. the Conversion of Arginine into Creatine in the Isolated Rabbit Heart

THE use of the whole animal for the study of the metabolism of creatine has a number of disadvantages. A large body of evidence exists which indicates that quite considerable quantities of creatinine may be injected into an animal, or administered parenterally, without the subsequent appearance of any extra creatine in the urine. Furthermore, the recent work of Benedict & Behre [1937] and of Ga...

Excretion of creatinine and creatine by beef steers.

In the classical theory of protein metabolism proposed by Folin (1) two types of protein metabolism are recognized, one a constant or endogenous metabolism, and the other a variable or exogenous metabolism. According to this theory the endogenous metabolism is represented by the excretion of creatinine at a relatively constant rate which is independent of protein intake. More recently, as point...

Guanidino compounds in guanidinoacetate methyltransferase deficiency, a new inborn error of creatine synthesis.

The first inborn error of creatine metabolism (guanidinoacetate methyltransferase [GAMT] deficiency) has recently been recognized in an infant with progressive extrapyramidal movement disorder. The diagnosis was established by creatine deficiency in the brain as detected by in vivo magnetic resonance spectroscopy and by defective GAMT activity and two mutant GAMT alleles in a liver biopsy. Here...

Creatine and creatinine metabolism in the normal male adult studied with the aid of isotopic nitrogen.

Experiments designed to demonstrate balance between the amount of creatine ingested and the amount retained or excreted as creatine and creatinine have so far failed in their purpose. The results of such studies have led to the opinion either that there is an end-product of creatine other than creatinine or that the synthesis of endogenous creatine is retarded by exogenous creatine (1). We have...
